Non competitive inhibitors are usually structurally unrelated to the substrate and bind to an allosteric site on the enzyme. Thus, both the substrate and the inhibitor theoretically can bind to the enzyme at the same time. The inhibitor however usually distorts the active site thus altering conformation of their catalytic residues.
